The Research Institute at Nationwide Children’s Hospital, Columbus, Ohio, in affiliation with The Ohio State University College of Medicine, Department of Pediatrics, has openings for post-doctoral fellows in the Patient-Centered Pediatric Research Program starting September 1, 2018. The Buck, a non-profit research institute, offers a rich scientific environment with renowned scientists in the field of aging research. The Buck is located 20 miles north of San Francisco & Berkeley, and is the only independent Institute in the U.S. devoted solely to research on aging and age-related diseases. Awarded a federal grant to establish interdisciplinary research in a new field called Geroscience, Buck scientists work in a unique, collaborative environment that allows scientists to initiate studies quickly and respond to new opportunities in fields such as stem cell research and regenerative medicine.


ABOUT THE CITP PROJECT:

The CITP project focuses on screening various compounds for their ability to extend the lifespan and “healthspan” of 21 isolate strains of three species of nematode worms in the genus Caenorhabditis. This animal model system is widely used in biomedical research. Goal 1 of the project is to test many different compounds to find the best candidates for ones that have potential to benefit people. Goal 2 is to focus on the reproducibility of results, achieved by doing the same experiments under tightly controlled conditions in all three institutions: University of Oregon, Buck Institute, and Rutgers University. The project data pipeline is idiosyncratic and highly customized. It is certain that unique challenges that cannot be specified or fully predicted will arise. The successful candidate will be able to show examples of ingenuity and resourcefulness in dealing with similar challenges.


JOB SUMMARY:

The Lithgow lab at the Buck Institute is seeking a Manager for a Data Coordination Center (DCC) for the three-institute consortium project, the Caenorhabditis Intervention Testing Program (CITP). The CITP aims to find robust and reproducible chemical interventions in aging by testing the effects of chemical compounds across many strains of the nematode Caenorhabditis in multiple laboratories across the country. This is a highly coordinated team effort requiring excellent communication skills.



KEY RESPONSIBILITIES:

Maintain and update the project data pipeline from assorted lifespan and healthspan experiments at the three institutions to the centralized Filemaker Pro 16 database.
Work with the PIs and technicians to simplify data quality control and metadata validation.
Ensure that data security and backup protocols are in place and tested routinely.
Work with project scientists to provide reports and output files for statistical analyses in third-party statistics packages.
Attend monthly project teleconferences and weekly technician teleconferences and address specific needs and project updates.
Maintain a “ticket list” of update requests and issue tracking.
Write or update existing scripts that allow smooth exporting and importing of data from one platform to another.
Work with the consortium institutes on any system-wide updates to software, operating systems, or specific experimental needs. We are testing a major upgrade to our Automated Lifespan system which requires updating our operating system to Scientific Linux 7.
Secondary needs include:
Investigate machine-learning improvements in the Automated Lifespan system
Look for more efficient methods to achieve project goals and reduce or eliminate sources of errors and duplicate effort


PROFESSIONAL REQUIREMENTS

BSc, MSc or PhD in the biological sciences
Experience working with scientific data in a multi-institutional or multi-department environment
Proven competence with Filemaker, MySQL, R and standard data formats in Excel
Excellent communication skills
Excellent time management skills requiring little direct supervision
An interest in the biology of aging, age-related diseases, or pharmacology

THE POSITION

The current position in the kapahi lab will involve modeling the influence of advanced glycation end-products in diabetes and neurodegeneration using C. elegans and mice The focus of the Kapahi Laboratory is to identify and characterize the mechanisms by which nutrients modulate aging and age-related diseases. This is being achieved by using an interdisciplinary approach combining genetic, pharmacological, biochemical and genomic approaches in invertebrate model systems C. elegans, D. melanogaster and mammalian cells. The broader significance of this research is to help uncover the role of nutrition in the etiology of age-related human diseases like diabetes, obesity, and neurodegeneration.
RESPONSIBILITIES

The current position involves developing models for diabetic complications and studying the effects of diet on aging and age-related diseases using C. elegans, mice and IPS derived cells. Postdoctoral position in mitochondrial physiology
Buck Institute
A postdoctoral position is available immediately in mitochondrial signaling in type 2 diabetes (T2D) in the Buck Institute for Research on Aging, Novato, California (in the San Francisco Bay Area).

This is a full time position with an initial one-year contract that can be extended.

The project uses pancreatic β-cell physiology assaying and top-down analysis of bioenergetic parameters measured using fluorescence microscopy and Seahorse respirometry to uncover causes of beta cell failure in T2D. Therefore both strong wet-bench and data analysis skills are required.

Project Overview
Altered cellular energetics and related cellular signaling contribute to human disease in general and physiological aging and chronic diseases in particular, including cancer, neurodegeneration, ischemic heart disease and type 2 diabetes. Mitochondrial metabolism also plays a central role in insulin secretion in pancreatic beta cells. When glucose is elevated, the increased metabolism results in a substantial mitochondrial membrane potential hyperpolarization, as well as in increased rates of ATP synthesis and turnover marked by faster cell respiration. The project uses microscale bioenergetic assaying of human primary β-cell cultures, assessment of β-cell physiology and top-down analysis of cellular energy metabolism. The project investigates the molecular pathways that make healthy β-cell bioenergetics – and insulin secretion – normally highly responsive to glucose, and pathways that may fail in type 2 diabetes. Relevant papers from our lab to this project are: PMIDs 26204464, 27404273, 27771512.

The successful candidate will receive training in the particular experimental technologies used in this project, will design and conduct experiments and analyze data. The postdoc is expected to lead his/her own project, write up and present results.

Essential Requirements:
PhD in biological or biomedical sciences, bioengineering or bioinformatics

Buck Institute Environment: The Buck Institute for Research on Aging consists of state-of-the-art research laboratories with faculty recruited from four different disciplines — diseases of aging, processes of aging, stem cell biology, and technology for aging studies. The labs were intentionally designed for close proximity interactions without departmental boundaries. Since its inception, the Buck Institute has collaborated closely with UCSF, UCB, LBNL and other San Francisco Bay Area researchers, clinicians and patient advocates at many levels. About 25% of Buck Institute faculty hold joint academic appointments with UCSF and/or UCB-LBNL. The Institute sits on 488 acres of Mt. Burdell in Marin County, California, 20 miles north of San Francisco. The long-term plans call for 550 employees in 45 faculty-supervised laboratories within five buildings totaling 360,000 square feet; to date, the first three buildings hold more than 300 employees in 16 laboratories and five cores. The facility has state of the art laboratories and support facilities designed and constructed with contemporary research methods and practices in mind. The Buck Institute is a multi-disciplinary facility and houses research associated with neurology, molecular genetics, molecular biology, cell biology, neurochemistry, cancer biology, mass spectrometry, proteomics, and apoptosis. Additional research support within the Buck Institute campus includes a 238-seat auditorium. The Buck Institute has developed several specialized multi-user core equipment facilities including the Morphology and Imaging Core (co-directed by Dr Akos Gerencser), a Bioinformatics and Genomics Core equipped with high- throughput microarray robotics, plate readers, laser capture microscope and multi-species genome probe libraries, as well as one of the Bay Area’s best equipped Chemical/Proteomics/Mass Spectrometry Core facilities.
