Postdoctoral fellow- gammaherpesvirus vaccine development

A postdoctoral position is available in the laboratory of Dr. Laurie Krug, HIV and AIDS Malignancy Branch (HAMB), Center for Cancer Research (CCR), National Cancer Institute (NCI)... , National Institutes of Health (NIH). The successful applicant will be part of a team focused on the developing a gammaherpesvirus vaccine. Our lab is dedicated to applying animal models of disease to understand viral and host determinants of chronic infection and pathogenesis by the gammaherpesviruses, Epstein-Barr virus and Kaposi sarcoma herpesvirus. The mission is to identify novel targets for intervention and prevention of human gammaherpesvirus-driven cancers.

Methods used in this research include a mouse model of vaccination and challenge, isolation of antibodies and immune cells from various tissues followed by neutralization assays, high resolution flow cytometry, microscopic imaging, and other state-of-the-art techniques. The project’s ultimate goal will be to translate these findings into human vaccine approaches. Our laboratory is part of a large program project involving many groups with broad ranging expertise, so the candidate will collaborate within and outside the Krug lab. The successful candidate should have a PhD or MD/PhD with a background in viral pathogenesis and/or immunology. Excellent written and verbal communication skills are expected.

Postdoctoral fellow: study how the kaposin RNA influences nuclear architecture during KSHV infection

The project will build off our recent work showing that the Kaposin RNA repositions nuclear speckles adjacent to the viral genome to optimize viral gene expression. There are many...  unanswered questions….such as 1) what does the 3D profile of the viral genome look like with/without kaposin 2) what is happening to cellular splicing factors localization/phosphorylation/function 3) what is the RNA feature that recruits speckles (structural or sequence) and 4) does the RNA need to remain bound to the viral DNA for this to occur? Take your pick!

This position offers an exciting opportunity for a talented researcher to investigate the role of virus encoded immunoregulatory proteins (e.g. Virokines) in the pathobiology of ILTV infections. Leveraging this knowledge, the project aims to develop innovative vaccination strategies, including cutting-edge vector-based and mRNA platforms, to combat ILTV. Fundamental experimental approaches of the laboratory include immunological, molecular, synthetic biology and cellular-based techniques along with a variety of animal models of pathogenesis and vaccine efficacy testing. Studies will be carried out in biosafety level BSL-2 and BSL-3 laboratories (if necessary). Successful applicants will be part of a diverse and multidisciplinary team focused on the experimental preclinical studies of avian infectious diseases. Additional research priorities include identifying, developing, implementing, and performing immunological analyses to assess the correlates of protection. We have developed an all-human three-dimensional (3D) SC/neuron co-culture system, amenable to the study of HCMV interactions. Our preliminary experiments have produced robust neu... rite outgrowth, SC/neuron interactions, MPZ expression and myelination. Initial experiments with pp71-expressing SCs produced very different interactions between the cell types and no evidence of myelination.
We propose to 1) Rigorously characterize our all-human in vitro SC/neuron 3D co-culture system designed to study HCMV's influence on MPZ production and myelination ; 2) Assess the ramifications of HCMV infection or sole pp71 expression on the baseline parameters established above; 3) Determine whether loss of MPZ function alone (via CRISPR KO in the SC) can replicate the perturbations we observe during infection- or sole pp71 expression and 4) utilize our co-culture system to examine crosstalk between the extracellular matrix (ECM) and myelin production, MPZ in particular, since the literature suggests a critical link between the ECM and SCs’ ability to properly myelinate neurons.

Applicants must have a Ph.D. in molecular/cellular biology/neurobiology, virology or a related field and a strong publication record in peer-reviewed English language journals. They must have extensive practical experience with standard molecular/cellular biological techniques and performance of tissue culture driven experiments. Experience in stem and neural cell culture, advanced molecular techniques (ChIPs, qPCR, quantitative realtime RT-PCR, CRISPR-driven experiments) and/or advanced microscopy techniques (confocal, TEM), is highly desirable.
