Recently, RNAs with complex glycans (glycoRNAs) have been identified in mammalian cells. These glycoRNAs are small non-coding RNAs. The role of these biological molecules is not known. In this project, the interdisciplinary team will design synthetic glycoRNAs that will be produced by the chemist members of the group. The selected candidate will aim to chemo-enzymatically synthesize glycoRNAs using standard techniques in synthetic chemistry and enzymatic biochemistry (e.g., performing thin-layer chromatography, using a rotary evaporator, analyzing an NMR spectrum or a mass spectrum, and using HPLC).

RNA’s simple chemical structure, composed of four nucleotides, contrasts with its highly complex functionality. Over 170 RNA modifications have been identified, impacting its func... tion and stability. The Höfer lab primarily focuses on characterizing RNAs modified at their 5' terminus with the redox factor nicotinamide adenine dinucleotide (NAD). Since the discovery of NAD-modified RNAs in 2015, they have been found in all kingdoms of life.
Building on these findings, the Höfer lab aims to understand the functional role of NAD-RNA in model organisms such as Escherichia coli and bacteriophage T4. The lab's research integrates synthetic biology, biochemistry, chemistry, and bioinformatics to uncover the connections between redox biology, gene expression, and regulation. Recent discoveries, including enzymes responsible for NAD-RNA biosynthesis and removal, offer insights into NAD-RNA's biological significance e.g. regulating RNA stability. In 2023, the Höfer lab introduced the concept of RNAylation—how RNAs and proteins interact—which provides a new perspective on RNA functionality (Wolfram-Schauerte, Nature 2023). However, the broader biological roles of NAD-RNAs remain poorly understood.
We look for candidates that would like to study the functional role on NAD-capped RNA in bacteria and bacteriophages or aim to apply the beauty of RNA-modifications in the field of synthetic biology.

The successful applicant should hold a degree in Molecular Biology, Biochemistry, or a related discipline.

The fellow will join a young, highly interactive, interdisciplinary and collaborative research team at the Max Planck Institute for Terrestrial Microbiology in Marburg, which is a world-leading hub of microbiology with state of the art research infrastructure and a highly stimulating intellectual environment.

Post-doc: Immune response to glycoRNAs in mouse model

Recently, RNAs with complex glycans (glycoRNAs) were discovered in mammalian cells. These glycosylated RNAs are small, non-coding RNAs. Little is known about the biological role... s of these molecules. In this interdisciplinary project, the impact of glycosylation on the immune response to synthetic RNAs will be investigated in a mouse model. GlycoRNAs will be designed and produced by chemists on the team. The person selected for this position will then be responsible for testing the immune response to these RNAs in mice using techniques such as multiparametric flow cytometry and RT-qPCR among others.

Candidate profile: Candidates should hold a Ph.D. in immunology or a related field. They should have extensive experience in the design and interpretation of flow cytometry experiments, and with murine experimental models, as demonstrated in publications.

We are a young lab trying to reconstruct the origins of RNA-based cellular life using a wide range of approaches including, but not restricted to, in vitro RNA evolution, structural biology, RNA enzymology, and synthetic cell research. We are also interested in understanding the fundamental mechanisms underlying RNA evolution. We are developing RNA-based tools to interrogate frontier problems in RNA biology such as transcriptomics of terminally-modified RNAs. Therefore, our lab straddles both extinct and extant RNA Worlds.

The project will employ multidisciplinary approaches in the areas of single-molecule and super-resolution microscopy, RNA/protein biochemistry, genomics and CRISPR-Cas9-mediated genome editing using mammalian cell culture models.
Our lab focuses on advancing the understanding of gene regulation by developing and applying single-cell and single-molecule approaches, combined with biochemistry, genomic, proteomic, and computational methods to fundamental questions in RNA biology (Adivarahan et al., Mol Cell 2018; Bensidoun et al., Mol Cell 2022; Jayabalan et al., 2021; PNAS; Lessard et al., Nature Cell Biology 2018). Building upon the Serratus project (www.serratus.io) we will explore the fringes of Earth’s Virome. We developed an open-source cloud computing architecture to analyze 5.7 million sequencing datasets (10.2 petabases); discovering >130,000 novel RNA viruses (only 15,000 were known previously)… in only 11 days. Learn more in our first Nature paper [nature.com/articles/s41586-021-04332-2].

Today we're continuing the search, pushing to discover 100 million RNA viruses by 2030, and prioritizing which ones are pathogens causing human disease.
